泌乳激素為腦垂腺前葉所分泌多肽荷爾蒙且在所有脊椎動物中具有多樣的生物功能。由於禽類開始孵卵行為 (藾菢) 受到泌乳激素增加而誘發,雞泌乳激素基因被認為是影響產蛋量之候選基因。先前的研究針對雞泌乳激素啟動子區域多型態與產蛋之關係做研究,多型態分析包括白來亨雞、海蘭褐蛋雞、肉雞與數種中國本地雞種。定序後發現在啟動子近端區域具有24個鹼基對之插入-缺失,在全啟動子區域也發現多個單核苷酸多型態而六個單核苷酸多型態基因型建構出八個單倍體。24鹼基對之插入純合子造成無藾菢,H3(C C T C T G)則是對產蛋最具優勢之單倍體。以即時定量聚合酶鏈鎖反應與放射免疫分析法研究24鹼基對與雞泌乳激素mRNA表現、血漿泌乳激素含量與藾菢行為之關係,結果發現基因型AB (插入/缺失) 具有藾菢行為最高發生率、最高雞泌乳激素mRNA含量,綜合以上結果說明此多型態可能藉由調控雞泌乳激素基因轉錄作用影響雞隻藾菢行為。

Title: Association of polymorphisms in the promoter region of chicken prolactin gene with broody traits and egg prodution
Abstract:
Prolatin (PRL) is a polypeptide hormone secreted by the anterior pituitary gland and have diverse biological functions in all vertebrates. Because the onset of poultry incubation behavior (broodiness) is induced by an increase in prolactin secretion, the chicken prolactin hormone plays a crucial role in broodiness. Hens cease egg production during the incubation period, so chicken prolactin gene is considered a physiological candidate gene for egg production. The current research was to investigate the association of polymorphisms in the chicken PRL promoter region with egg production. For polymorphism analysis, White Leghorns, Hy-Line brown egg layers, broiler, and many Chinese local breeds were included. Sequencing revealed a 24-bp indel (insertion-deletion) occurring in the proximal promoter region, several single nucleotide polymorphisms (SNPs) were also found in promoter and 6 SNPs genotype constructed 8 haplotypes. Homozygous insertion of the 24-bp sequence lead to nonbroodiness, H3 (C C T C T G) was the most advantageous haplotype for egg production. Real-time quantitative PCR and radioimmunology assay were used to investigate the potential association of the 24-bp indel locus with chicken prolactin (cPRL) mRNA expression, plasma cPRL and brooding behaviors. The results showed that chickens with genotype AB (insertion/deletion), which are the highest incidence of broodiness, had the highest cPRL mRNA level. It is concluded that this polymorphic site might be related to the broodiness in chickens via modulating the transcriptional level of cPRL gene.
